EPIDERMAL KERATINOCYTES EXPRESS IL-13 mRNA AND ITS EXPRESSION INDUCED BY UVB IRRADIATION PRECEDES THAT OF IL-10 mRNA.

Epidermal keratinocytes play an important role in cutaneous immunologic reactions through the production of various kinds of cytokines. Especially, IL-10 has a great potency to suppress DTH response. IL-13 enhances IgE synthesis by B cells and suppresses the expression of inflammatory cytokines of monocytes. In this study, we detected IL-13 mRNA expression by cultured epidermal keratinocytes obtained from normal skin, using RT-PCR method. The expression of both IL-13 and IL-10 mRNAs were enhanced by UVB irradiation (UVR). IL-13 mRNA reached the maximum level 6 hours after UVR, whereas the maximum level of IL-10 was obtained at 12 hours. To our knowledge, this is the first report on the IL-13 expression by epidermal keratinocytes. These results suggest that IL-13 is an early anti-inflammatory cytokine derived from epidermal keratinocytes after UVR .

PRODUCTION OF IL-13 BY PERIPHERAL BLOOD MONONUCLEAR CELLS FROM PATIENTS WITH ATOPIC DERMATITIS.

To determine whether the production of IL-13 by peripheral blood mononuclear cells¡ÊPBMC¡Ëfrom patients with atopic dermatitis¡ÊAD¡Ëis elevated in AD, we examined PBMC from 52 patients with AD and 23 healthy volunteers as controls. PBMC were cultured either with or without the antigen extracted from Dermatophagoides farinae or housedust for 48 h, and then the supernatants were measured for IL-13 by competitive enzyme immunoassay¡ÊEIA¡Ë. We also examined the effect of immunosuppressant FK506 to the production of IL-13 by the antigen stimulated PBMC from patients with AD. Production of IL-13 by non-stimulated PBMC in AD were significantly higher than that in controls. The IL-13 levels in AD were increased after the antigen stimulation, and were higher than those in controls. The enhanced production of IL-13 in AD was suppuressed by FK506. These observations suggest that IL-13 may play some roles for the development of AD.

INTERLEUKIN-13 IN ATOPIC DERMATITIS.

Interleukin-13 (IL-13) is a novel cytokine which shares several properties with IL-4, including induction of IgE synthesis. Using RTŽ¥PCR, in situ RT and immunohistochemistry, we investigated the expression of IL-13 in the involved and uninvolved skin, and peripheral blood mononuclear cells (PBMC) of patients with atopic dermatitis (AD). We also examined normal skin and PBMC of healthy volunteers. By RT-PCR, IL-13 gene expression was detected in the involved and uninvolved skin and in PBMC of AD patients, but not in normal skin and PBMC of the healthy volunteers. However, by in situ RT, IL-13 gene expression was mainly detected in the involved skin. IL-13 protein was detected in the involved skin of AD patients and slightly expressed in the uninvolving skin of AD patients. These results suggest that IL-13 may play a role in the pathophysiology of AD.

depleting CD4+ T cells. Moreover, systemic administration of neutralizing Ab to IL-5 on day 0 completely inhibited the development of blood and tissue eosinophilia in vivo. Thus, CY pretreatment of PCl-sensitized mice probably allows the proliferation of T cells capable of producing IL-5 in the afferent phase, that results in eosinophil production and blood eosinophilia.¡¡

INFLUENCES OF RESPIRATORY ATOPY ON DISTRIBUTION OF EOSINOPHILS AND DEPOSITION OF EOSINOPHIL-GRANULE MAJOR BASIC PROTEIN IN ACUTE LESIONS OF ATOPIC DERMATITIS.

Although various authors have stated that infiltration of eosinophils plays an important role in pathogenesis of atopic dermatitis(AD), eosinophils are not always observed in skin lesion of AD. To see what the factors might be that are related to the infiltration of eosinophils in AD skin lesions, we ¡¡analyzed tissues immunohistologically for the presence of eosinophil-granule major basic protein(MBP). We took biopsy specimens from the acute lesions of 30 consecutive patients with AD, and stained those with anti human MBP antibody . There was a wide individual variation in the distribution of eosinophils and the dermal deposition of MBP in skin lesions of AD. A marked increase in both tissue eosinophils and dermal MBP deposition was seen in some patients with AD who had respiratory atopy.
